疏风解毒胶囊治疗新型冠状病毒肺炎的网络药理学研究

摘要:

目的:利用网络药理学探讨疏风解毒胶囊治疗新型冠状病毒肺炎的机制。方法:通过TCMSP网络平台检索疏风解毒胶囊的活性成分以及作用靶点;通过GeneCards数据库获取COVID-19的相关靶标。由Cytoscape软件和STRING数据库建立药物-化合物-靶标网络、靶标蛋白互作PPI网络。R语言的cluster Profiler工具包对于核心靶点展开GO功能富集以及KEGG通路富集分析。结果:根据筛选条件得到207个化合物和248个药物相应靶点。PPI核心网络包含48个蛋白,关键蛋白涉及MAPK1、MAPK3、STAT1、IL6等。GO功能富集分析取得1 618个条目(P<0.05),其中生物过程(BP)1 498个,细胞组成(CC)33个,分子功能(MF)87个;KEGG通路富集有157条通路与COVID-19相关(P<0.05),涉及人类巨细胞病毒感染、IL-17信号通路、HIF-1信号通路、TNF信号通路等。结论:疏风解毒胶囊中的活性化合物能通过多靶点、多通路治疗新型冠状病毒肺炎。

Abstract:

Objective: To explore the mechanism of Shufeng jiedu capsules in the treatment of Corona Virus Disease 2019(COVID-19)based on network pharmacology.Methods: The active compounds and targets of Shufeng jiedu capsules were searched through TCMSP network platform; The targets related to COVID-19 were screened by GeneCards database.The drugs-compounds-targets network and target protein interaction PPI network were constructed through Cytoscape software and STRING database.GO enrichment and KEGG pathway annotation analysis of core targets were performed using cluster Profiler toolkit for R language.Results: According to the screening conditions, 207 compounds and 248 drug targets were obtained.PPI core network contained 48 proteins, the key proteins included MAPK1,MAPK3,STAT1,IL6,etc.GO functional enrichment analysis had 1 618 GO items(P<0.05),including 1 498 biological processes(BP),33 cellular components(CC),87 molecular functions(MF).KEGG pathway enrichment analysis obtained 157 pathways related to COVID-19(P<0.05),involving human cytomegalovirus infection, IL-17 signaling pathway, HIF-1 signaling pathway, TNF signaling pathway, etc.Conclusion: The active compounds in Shufeng jiedu capsules can treat the COVID-19 by multiple targets and multiple pathways.
