目的:研究采收月份对新疆紫草质量的影响,为合理确定新疆紫草药材采收时间提供依据。方法:以4-9月份下旬采集的新疆紫草根为材料,称量鲜重和干重,采用UV、HPLC测定羟基萘醌总色素、 β,β′-二甲基丙烯酰阿卡宁的含量,通过UHPLC-MS测定代谢产物,采用ABTS、DPPH、FRAP法检测体外抗氧化活性,评价对黑色素瘤细胞A375的抑制作用。结果:新疆紫草生物量呈先降后增的趋势,9月份最高;2个指标成分呈先增后降再增的趋势,且分别在5月和9月含量最高;中药非靶向代谢组共检测到1 291个代谢产物,其中578个差异代谢产物在5月高表达,其次是9月。体外抗氧化结果显示5月份的抗氧化能力最佳。体外抗肿瘤细胞实验结果表明5、8、9月样品对A375细胞增殖有一定的抑制作用。结论:该研究阐明不同采收月份新疆紫草生物量、指标性成分含量、代谢产物、体外抗氧化试验及抗肿瘤活性的变化规律,为新疆紫草的科学采收提供了依据。

Abstract:

Objective:To study the influence of the harvest month on the quality of Arnebia euchroma,so as to provide a basis for determining the optimal harvest time of Arnebia euchroma. Methods:The roots of Arnebia euchroma plants collected in the latter part of each month from April to September were used as materials,the fresh weight and dry weight were measured,and the contents of the total hydroxynaphthoquinone pigments and β,β′-dimethylacrylalkannin were determined using UV and HPLC methods,respectively. Metabolites were analyzed by UHPLC-MS,and in vitro antioxidant activity was detected using ABTS,DPPH,FRAP assays. Additionally,the inhibitory effect on melanoma A375 cells was assessed. Results:The biomass of Arnebia euchroma initially decreased and then increased,reaching its peak in September. The two indicator components exhibited a trend of initial increase,followed by a decrease,and then another increase,with the highest contents observed in May and September,respectively. Untargeted metabolomics analysis detected 1 291 metabolites,among which 578 differential metabolites were highly expressed in May,followed by September. The in vitro antioxidant results indicated that the samples harvested in May exhibited the strongest antioxidant capacity. The in vitro antitumor cell experiments demonstrated that samples collected in May,August,September had certain inhibitory effects on the proliferation of A375 cells.Conclusion:This study elucidates the variations in biomass,indicator component content,metabolites,in vitro antioxidant activity,and antitumor activity of Arnebia euchroma across different harvesting months,providing a scientific basis for the optimal harvesting of Arnebia euchroma.
